4. Outbreak of replication-competent retrovirus in "Helper-Free" Unigenomic Lines

 

In the unigenomic lines there is a potential for outbreak of helper virus by recombination of the introduced retroviral construct and resident packaging constructs. The fewer the number of recombination events that are needed the more likely thepossibility is that a wild-type replication-competent virus can arise (and ruin your experiment, or in the case of an amphotropic break-out, ruin more than that...)
