My research aims at
elucidating the mechanism of the copper containing enzymes Nitrite
Reductase (NIR) and Nitrous Oxide Reductase (N2OR),
which
catalyze the second and last step respectively, of the bacterial
denitrification cycle. I am also studying the mechanism of cofactor
biogenesis of topaquinone in mononuclear copper Amine Oxidase, and
related model complexes.
I
use several spectroscopic techniques such as
Absorption, CD, MCD,
resonance
Raman, EPR and DFT calculations to understand the ground
state wave function of these enzymes and related model complexes. I
also perform parallel kinetic studies using stopped flow kinetics and
rapid freeze quench techniques to elucidate the steps involved in the
enzymatic turnover.
